About Me We investigated the effect of n-tert-butyl-alpha-phenylnitrone (pbn) spin free radical scavenging on focal cerebral ischemia/reperfusion injury in rats anesthetized with halothane. 90 min after occlusion of the middle cerebral artery with an intraluminal thread, the artery was reperfused. Intravenous injections of 25 mg/kg pbn five minutes before and half an hour after the introduction of the thread significantly reduced the volume of the lesion, measured one day after ischemia. During ischemia and during the first 30 min after reperfusion, cerebral blood volume and blood flow were measured using volume-sensitive and recently developed flow-sensitive magnetic resonance imaging (mri) and using laser doppler flowmetry. In different animals, the area of decrease in blood flow exceeded the area of decrease in volume by two, twice. The area of post-reperfusion blood flow deficit corresponded to the final 24-hour lesion volumes measured histologically, much more effectively than limiting the amount of blood in animals treated with saline and pbn. Reperfusion resulted in return of blood flow and complexity to values close to the contralateral side in the pbn-treated animals, not that in the saline-treated control group. We will calculate that during focal ischemia/reperfusion, pbn gives protection to the vascular endothelium, leading to an increase in postischemic reperfusion.
